Based on fold-change analysis and statistical analysis, we identified 16 up-regulated miRNAs (mir-240, mir-75, mir-787, mir-62, mir-251, mir-252, mir-1821, mir-360, mir-353, mir-254, mir-229, mir-1824, mir-795, mir-1820, mir-41, and mir-4923b) and 4 down-regulated miRNAs (mir-4812, mir-53, mir-794, and mir-86) in nematodes infected with C. albicans SC5314 (Figs 1, 2a,b and 9, Table S2). The up-regulated miRNAs included mir-240, mir-75, mir-787, mir-62, mir-251, mir-252, mir-1821, mir-360, mir-353, mir-254, mir-229, mir-1824, mir-795, mir-1820, mir-41, and mir-4923b, while the down-regulated miRNAs included mir-4812, mir-53, mir-794, and mir-86 in C. albicans SC5314 infected nematodes (Fig. 2a,b). We first tested the ability of TRM mutant adults to lay eggs in a 24-hr period at 20°C and found that loss of mir-229 mir-64-66 caused a slight drop in egg-laying rate (Figure 4A), suggesting a role for these miRNAs in the regulation of egg-laying. [score:2]
Taken together, these observations show that mir-71 and mir-229 mir-64-66 mutant animals are short-lived at 25°C in addition to being heat stress sensitive. [score:1]
In these experiments, we confirmed that mir-71 and mir-229 mir-64-66 are required for normal lifespan at 25°C as they are at 20°C. [score:1]

First, we found that the brood sizes of TRM mutants were not significantly different to wild type animals at 20°C, with the exception of mir-229 mir-64-66 which has a small decrease in brood size (Figure 4E). [score:1]
We observed that loss of mir-71, mir-229 mir-64-66 and mir-239 caused significant decrease in brood size compared to wild type animals at 25°C, yet caused no embryonic lethality (Figure 4F and data not shown), suggesting that these miRNAs are required to regulate gamete production in high environmental temperatures.
