To further decipher the functions of endogenous dTau proteins in Drosophila, we performed homologous recombination to generate a tau knock-out (KO) line presenting a selective deletion of the E2 to E6 region of the tau gene (exon numbering based on FlyBase data for tau-RA transcript), hence avoiding mutation of surrounding genes such as rps10a and mir-1001 (Fig. 2a).
