Five miRNAs (miR169, miR172, miR397, miR398, and miR827) were identified as being differentially expressed in leaves in response to the transient low N condition, with miR172 up-regulated but miR397, miR398, and miR827 down-regulated (Table S1). [score:9]
Under N-limiting condition, three of the miRNAs (miR164, miR172, and miR827) were up-regulated while the others were down-regulated (Table S1). [score:7]
The Arabidopsis miR827 is specifically up-regulated by phosphate deficiency [16], where the expression level of miR827 didn't show significant change to N limitation [37]. [score:6]

Three of these five miRNAs (miR172, miR397, miR398) shared the same expression patterns in their response to both chronic and transient N-limiting conditions, while one of them (miRNA827) showed an inverse response to the two conditions (Table S1, Fig. 1). [score:3]
With regards to tissue specificity(or tissue dependent), some miRNAs were only regulated in roots or leaves, such as miR160, miR167, miR168, miR319 and miR395 in roots, and miR164, miR172, miR397, miR398 and miR827 in leaves, while some others were regulated in both tissues, such as miR169, miR399, miR408 and miR528 (Fig. 4). Nine miRNA faimlies (miR164, miR169, miR172, miR397, miR398, miR399, miR408, miR528, and miR827) were identified to be differentially expressed in leaves in response to chronic low N condition. [score:3]
The second category includes miR395, miR397, miR398, miR399, miR408, miR528, and miR827, whose potential target genes are predominantly involved in energy metabolism and scavenging of the oxidative species produced during stress. [score:3]
The characterization of rice osa-miR827 and its two target genes, OsSPX-MFS1 and OsSPX-MFS2, provided evidence that they may target different genes compared with Arabidopsis and play a role in phosphate (Pi) metabolism [37]. [score:2]

We also discovered several miRNAs that were either downregulated or upregulated by N limitation, including the conserved miR169s, miR395s, miR528s, and miR827 and the newly identified miRC1, miR169w, miRC19, miRC37 and miR171p/q. [score:7]
As illustrated in Figure 6, the expression of miR169, miR395, miR827 and miRC1 was down-regulated by N-limitation and formed the first subgroup. [score:6]
MIR162 and MIR167s were also upregulated by N deficiency in roots, and the expressions of MIR169s, MIR397s, MIR408s, MIR528s, MIR395s, and MIR827 were clearly decreased in N -deficient roots (Figure 3B). [score:6]

In our deep sequencing data, several miRNA* (miR159d*, miR169m*, miR171h*/k*, miR319b*/d* and miR395o*) showed a greater number of normalized small RNA reads than the corresponding mature miRNA, and we even validated the potential targets of miR164f*, miR167g*/h*/i*, miR169r*, miR168a*/b* and miR827* by the degradome (Table 3), in line with previous findings from vertebrate [46] and Medicago truncatula [47]. Because of the importance of roots in the sensing and uptake of nutrients, we selected MIR169c/r, MIR395a/b/d/e/f/g/h/i/j/n/p, MIR528a/b, and MIR827 for further validation by Real-time RT PCR or small RNA northern analysis (Figure 3C and D). [score:1]
Though recent evidence showed that miR827 took part in maintaining nitrate -dependent phosphate homeostasis in Arabidopsis [49], the function of miR827 in plant adaption to N limitation requires further research. [score:1]

Moreover it is predicted to target an mRNA encoding a sec14 cytosolic factor [29] while both osa-miR827a and ath-miR827 target an mRNA encoding an SPX protein. [score:5]
It has also been shown that the predicted SPX target of ath-miR827 is cleaved in vivo at the predicted miRNA complementary site [7]. [score:3]
Significantly, both the rice and Arabidopsis miR827 are predicted to direct cleavage of mRNAs encoding proteins with an SPX motif. [score:2]
These data strongly suggest a role of osa-miR827a and ath-miR827 in the response of rice and Arabidopsis to phosphate or nutrient availability. [score:1]
This miRNA is the homolog of recently reported ath-miR827 but differs by two nucleotides [7]. [score:1]
